The two players were downgraded on Saturday after previously being listed as doubtful.
Matthews aggravated an ankle injury in the 17-14 loss at the Minnesota Vikings last Sunday night. He also is dealing with a hamstring injury.
Burnett sustained a hamstring injury in the second half against the Vikings, but he was listed on the injury report this week with a groin injury.
The Packers also downgraded outside linebacker Datone Jones (knee) and defensive tackle Letroy Guion (knee) from questionable to doubtful.
Jones did not practice this week and Guion is recovering from a sprained MCL he sustained in the second quarter against Minnesota.
Packers cornerback Sam Shields was already ruled out with a concussion.
The Lions will be without their best pass rusher, defensive end Ezekiel "Ziggy" Ansah, who was ruled out Friday with an ankle injury.
Ansah injured the ankle during the first series of Detroit's loss to the Tennessee Titans last Sunday.
The Lions are likely to be without linebacker DeAndre Levy, who is listed as doubtful, for the second straight week. Levy, who is listed as doubtful, has missed every practice the past two weeks with a quad injury.
In addition, the Lions listed another key defensive player, linebacker Antwione Williams, as doubtful with a hamstring injury.
