The two-hour star-studded special set to air live on the network on Thursday at 9 p.m. ET, will help raise money and awareness for children living in poverty.
"Red Nose Day is on a mission to help lift kids out of poverty," a press release explained. "People from all walks of life are encouraged to get involved, have fun and raise money by buying red noses sold exclusively at Walgreens stores nationwide, organizing fundraising events and watching and donating during the television special."
The new collection of musical gusts will be joined by a long-list of celebrities including Ellen DeGeneres, Jack Black, Paul Rudd ,Ludacris, Zac Efron, Julianne Moore, Liam Neeson, Sarah Silverman, Tracy Morgan, Key & Peele, Anna Kendrick, Seth Rogen, Ellie Kemper, Kristen Bell, Emma Watson, Rose Byrne, Blake Griffin, Emma Thompson, Chloë Grace Moretz, Steve Buscemi, Danny Trejo, Adam Devine and Ron Funches.
Hosted by Craig Ferguson, the telethon will feature musical performances and comedic short films. In a preview for the event, Kendrick was seen as a female version of Indiana Jones as she hilariously attempted to recover the fabled holy grail, hinting at what's to come.
Stars such as Julia Roberts, Pharrell, Will Ferrell, Michelle Rodriguez, Shelton, Reese Witherspoon, Black and Ed Sheeran took part in last year's show, which helped raise more than $23 million for the Red Nose Day Fund.
