The Jayhawks built a 40-12 advantage while handing Towson its 20th consecutive loss.
Kansas hit 59 percent of its shots from the field and forced 23 turnovers. The Jayhawks turned the ball over just five times.
The game was the first as Towson coach for former Pittsburgh assistant Pat Skerry. He replaced Pat Kennedy after Towson went 4-26 last season.
Kansas will next meet No. 2 Kentucky Tuesday in New York.
