The fireworks from Doumit ended the Tigers' four-game winning streak and cut their lead over the Chicago White Sox in the American League Central to a single game.
The teams battled to a scoreless tie through six innings before reliever Phil Coke gave up Doumit's 17th homer of the season. That blast came after Justin Morneau had reached on a fielding error by Detroit first baseman Prince Fielder.
Minnesota third baseman Trevor Plouffe then committed a fielding error in the Detroit eighth that allowed Austin Jackson to reach base and Omar Infane followed with a home run that tied the game.
The Twins jumped back in front in the bottom of the eighth when Brayan Villarreal (3-5) walked the bases loaded, one of them intentional, and gave up Doumit's two-run double.
Doumit has 71 RBI this season, a career best.
