Conklin stopped all 26 shots fired his way to get his first shutout since Jan. 3, 2006, with Edmonton against Chicago.
Pittsburgh's goals were scored by Colby Armstrong in the second and Evgeni Malkin in the third, after assists by Armstrong and Sidney Crosby.
Malkin's 15th goal this season trickled into an empty net with 22 seconds remaining.
Ryan Miller was beaten just once on 24 shots for the Sabres, who have lost three in a row after a six-game winning streak.
It was a costly victory for Pittsburgh, which lost forward Gary Roberts to a broken left leg in the second period. The injury occurred after Roberts got tangled up with Buffalo's Tim Connolly in the left circle and both landed awkwardly.
The Penguins are uncertain how long Roberts will be sidelined.
